The vagina isn't an open hole that has a certain diameter. It's all shut up unless you put something in there -- a tampon (small), a penis (hopefully larger) or a baby comes out (kinda scary big). The external genetalia obviously comes in great variety and can create a varience in the opening to the vagina I suppose, but once you get in the vagina itself closes around whatever goes in or out. What also leads to variety is muscle tone -- the PC muscle will make a looser or tighter grip.
As for virginity, if the hymen is still intact, that will be another 'obstacle' at the opening of the vagina. Also it will be the first time it is stretched that big, never having had something that size put in it before, so it sort adjusts, not changes.
