3,4-diaminopyridine is a drug that:
* increases acetycholine release from the nerve terminals
* blocks potassium conductance, prolonging the action potential at the nerve terminal, and increasing acetylcholine release.
* initial dose is usually 10 mg, four times a day.
* side effects include:
o headache
o fatigue
o perioral and/or distal paraesthesia 30-60 minutes after a dose
o difficulty in sleeping
o excessive dosage can lead to seizures
* it is contraindicated in epilepsy
the trial was not double blinded so the result is highly in question. toxicity studies also call into question the cost benefit of the drug as 11% of patients had severe adverse reactions
