Depends upon what you mean.
The urine tests are testing for the presence of certain substances. If EITHER of the two people who urinated into the sample had that substance in them, then the test will detect it.
For example, if either were a heavy marijuana smoker, then marijuana would show up for 30 days in their urine.
But otherwise, no - there will be no "failed" test if two people urinated into the same cup for the test. If both people were "clean", then their urine would be nearly identical. With all basic urine testing, there would be no way to even determine if two people urinated into the cup.
Expensive DNA testing on a few possible cells from the urinary tract could definitively determine that 2 people likely did pee into the same cup...but no one would ever do that, except for a crime investigation, etc.
In the end, tap water should have been used to "fill up what was needed"...and all would have been well. The vast majority of urine is simply water, anyway...and unless you add a LOT of water, tests would never know.
This is why for an "official" urine drug screen, a dye is poured into the toilet, so the person can't just dip the cup in the toilet. And, tap water couldn't be exclusively used...of course.
As it is, if the person "scamming" to hide the presence of cocaine, marijuana, etc. from their own urine by using the urine of another person...didn't get enough urine from their friend...and then urinated themselves to fill it up, then yes...they will likely get caught.
