Answers:
When you have knee pain, it can be due to many factors. In your case, if you had either a calcium deficiency or overdose, then in either case, you'd have symptoms all over and not localized to just your knees. Sitting for long periods of time will cause aches and pains and keeping any joint bent in the same position will cause pain. Lack of physical exercise, being sedentary, or obese and sedentary will cause that sensation of muscle weakness when walking or exercising. Take frequent "body position breaks" during your work hours. Learn to tap your feet or circle your ankles, if you can't get up frequently enough to relieve the pain. If none of these solutions work, then go get checked up by your Doctor.

I think it should be that you are sitting too much. Take brisk walks every half an hour to stretch you muscles. A walk to the kitchen for a snack or a cup of water is good enough.

Calcium imbalance produces primarily muscular problems. Could be arthritis, but at your age, I doubt it. Any injuries, old or recent? weakness could be a sign of more serious things like a muscular dystrophy, but the knee pain wouldn't be your primary complaint.
