Yes. You can do it! just takes a bit more work & diet.
The short answer is, of course. The long answer is your question leads me to believe that you have tried and failed to get benefit from weight training or other exercise and wonder if that is because you are 40+ years old.
It's possible. Many people, as they get close to middle age start to become deficient in growth hormone. This causes various symptoms..depression, lack of libido, impotence, a general degradation in physical fitness and a failure to respond to exercise.
If this is you, what you should do is make an appointment with an endocrinologist. Find one that specializes in growth hormone supplementation. He should pull a hormone panel on you to see if in fact you are deficient. If that's the problem (and it is a very common problem), he will precscribe injections of growth hormone (the exact amount needed to bring your levels back up to normal..no more). This should fix the problem. Your symptoms should disappear and you will get normal results from exercise again.
