Yes, there are diseases that cause people to gain weight. A former co-worker of mine has a disease that causes her to go through some weight-gaining periods every now and then and causes her fatigue as well. She has to take medicine regularly for it to keep it under control. To the best of my knowledge, there are other diseases that might do the same thing. I would suggest that she visit a doctor about this. It's not that I think it is a life-threatening disease, just lifestyle threatening.
There are not many diseases that cause overweight, but there could be a thyroid problem. Also there can be certain medications that cause weight gain such as certain anti-depression meds. Also if there is any injuries in the bones that would cause pain it would be harder to exercise. So I guess you know her more than we do and you can analyze from there :)

Also, forgot about PCOS. She could gain up to 100 pounds from that alone. Your mom needs some bloodwork drawn to make sure everything is ok.
