Monte gave a great answer but there are a couple of more things that it does to help you. Number one because of the added weight, it provides resistance and resistance builds muscle and strength. In other words it will help increase muscle and strength in your legs. Also track athletes and football players at speed positions use weight vests sometimes because it will also build speed. If you train with a weight vest properly you will find that your times in sprints and the 40 will get faster.

you simply burn more calories than you would usually. The weight is more distributed around your core and shoulders so it isn't as straining as hand and ankle weights.
It adds more weight which will provide more resistance and will burn more calories.
